A PAPS-Dependent Sulfotransferase in Cyanophora paradoxa
نویسنده
چکیده
A 3'-phosphoadenosine-5'-phosphosulfate (PAPS) dependent sulfotransferase was partly purified from Cyanophora paradoxa. This enzyme has a pH-optimum of 8.5 and was found to be specific for P A P S ; the K m for this nucleotide was determined to be 8.3 juu. In crude extracts monothiols like glutathione, mercaptoethanol, or cysteine catalyzed a rate of about 80% of the rate obtained with dithioerythritol; however after purification of the PAPS-sulfotransferase the monothiol activity decreased to about 20% of the rate obtained with dithiols. Addition of heated, crude extract partly restored the monothiol activity. This PAPS-sulfotransferase was inhibited by 5'-ADP and 5'-AMP, and especially by adenosine-5'-phosphosulfate (APS). Practically no inhibition was found with 2'-AMP, 3'-AMP, c-AMP, 3'-5'-ADP and 2'-5'-ADP. The inhibitor constants have been determined to be 430 /uu for 5'-AM P; 290 /um for 5'-ADP; and 0.3 f iu for APS. These inhibitions seem to be competetive to PAPS. The role of these inhibitors for a possible regulation of assimi latory sulfate reduction are discussed.

Distribution of thioredoxins in Cyanobacteria.
The presence of thioredoxin was demonstrated in 20 strains of cyanobacteria as well as in one phototrophic bacterium Rhodopseudomonas sulfidophila and in Thiobacillus denitrificans. Thioredoxin activity was not found in Cyanophora paradoxa and in Porphyridium cruentum using the thioredoxin-dependent PAPS-sulfotransferase activity from Synechococcus 6301 as assay system.
متن کاملCyanophora paradoxa: Fatty Acids and Fatty Acid Synthesis in vitro
Cyanophora paradoxa, Cyanelles, Fatty Acid Pattern, Fatty Acid Synthesis in vitro The fatty acid pattern of Cyanophora paradoxa membrane lipids is highly unusual with 16:0, 20:3 and 20:4 as the main acids. The 20:4 acid is preferentially distributed among the cyanelle lipids. In isolated cyanelles a relatively low in vitro synthesis of only saturated and monounsatu rated fatty acids from [l-14...

Culture conditions are described under which Cyanophora paradoxa grows with a generation time of less than two days. The organism is an obligate photoautotrophic flagellate unable to degrade exogenous carbohydrates via respiration. It does not synthesize nitrogenase but can form a hydrogenase with similar properties as in blue-green algae. The photosynthetic 0 2-evolution pro ceeds with essent...
متن کاملPathway of cytosolic starch synthesis in the model glaucophyte Cyanophora paradoxa.
The nature of the cytoplasmic pathway of starch biosynthesis was investigated in the model glaucophyte Cyanophora paradoxa. The storage polysaccharide granules are shown to be composed of both amylose and amylopectin fractions, with a chain length distribution and crystalline organization similar to those of green algae and land plant starch. A preliminary characterization of the starch pathway...
متن کاملRibonuclease-P RNA gene of the plastid chromosome from Cyanophora paradoxa.
The gene, rnpB, encoding the RNA portion of ribonuclease-P has been found in the cyanelle DNA of Cyanophora paradoxa. A secondary structure model for the cyanelle RNA fits into that for eubacterial Rapb-RNAs.
